Taking the tape off to hone with just water shouldn't affect anything, but it's not going to help anything either. No reason to do it - all it does is take off a minute amount of steel from the back...

If you set the bevel with tape, you need to keep the tape on for the rest of your honing. Without the tape, you'll just be honing the back of the bevel, not the edge itself.
