I don't read very many but this one is by far the best I've found.
Lackadaisy
Its about gangster cats during prohibition and the speakeasy their lives revolve around. Fantastic sepia-tone art and a great storyline.
Its not updated as frequently as I like but the art alone looks like it takes a lot of time.
What other strips should I be looking out for?

One of my favourites is xkcd - A Webcomic - Sierpinski Valentine but that's probably because of my geeky sensibilities. I just love this one.
If you like dark/ironic humour, The Perry Bible Fellowship is great. Though he released a hardcover of his comics a year or two ago and really hasn't had anything new since.

A comic a day. Often thought provoking and educational, and if you miss a few days you can usually read the new eipisodes in reverse order without ruining it (the theme changes frequently.)
Beware, he may direct to you the tvtropes website and you will lose hours.

Evil Inc. by Brad Guigar - Daily Super-Villain Webcomic and Comics Blog
Evil Inc. is a fantastic comic about heroes and villains that typically gets a chuckle out of me. The story lines are actually very good, and I love to get each new one.
Penny Arcade! - New York Sketchbook: A Nose Journey
Hilarious observational comics on the world of geek, focusing on video games, but really running the gamut. These are also the guys that run www.childsplaycharity.org
I've got 'em both on my topside Firefox RSS feed and read them both daily.
