I converted "my" HPLC into a nanoLC and it's now (in theory) 400x more sensitive! I can detect tiny tiny amounts that previously disappeared in the machine's noise.

For those who want to know, a short explanation:
HPLC is short for high performance liquid chromatography, a separation method that I'm currently using a lot to analyse snake venom. It helps separate the different proteins in snake venom.
NanoLC is a variant that uses slightly different equipment, which allows for much lower sample requirements and much better detection.

Will this help you to make synthetic netherparts incase your netherparts get seat posted? Does it use the same equipment, or did you have to replace a lot of it?

Just curious, if we stay off topic doesnt that mean we are staying on topic, but if we stayed on topic we'd be off topic, based on the topic.

Yes and no, if we have absolutely no conversations, just random bullet points then the thread will just disolve into dribble, i dont see a problem with conversations etc, i did set an arbitary 3 post limit for exchanges, but ultimately this thread is for those of us who feel a need to talk about nothing to come to and do so. I like the fact that we can do that, and reanimate conversations that may have stopped 100 posts ago, or just post something fun or silly. Its sort of like the kitchen at a party if you will.

You can also cross post to here if you like, you know, quote a post from a different thread and make comments etc that might not be germain to the conversation there, you might then need to post a link to this thread in that one, but maybe we can do a service to the forum with our bold endevours

Not at all The closest related thing I can say is that I'm now working with a 20 cm column.

As for the equipment, I'm still using the same high pressure pump and mass spectrometer (which measures the weight of the molecules in your mixture and then you have to "guess" what that weight means). The main replacement was from a separation column of 2 mm diameter to a 0.1 mm diameter. Taking into account that the columns relate as 22:0.12 = 4:0.01, the sensitivity should have increased about fourhundredfold (haven't measured the exact increase, but it sure is a lot better).

I like explaining these things! The separation of a mixture with HPLC is based on pumping the mixture through a column with, for instance, hydrophobic material. Hydrophilic molecules will travel faster than hydrophobic ones, hence the separation
